PCR vs. Culture Comparison

1. Polymicrobial Detection: PCR offers a more effective approach for identifying bacteria in chronic wounds, which often contain multiple bacterial species, compared to standard culturing methods.

2. Speed Advantage: Whereas traditional culturing can take 3-5 days, PCR provides results in 24-48 hours, enabling faster clinical decisions.

3. Increased Sensitivity: Standard lab culturing may miss up to 84% of clinically relevant bacteria in chronic wound samples, highlighting PCR’s superior sensitivity.

Bacterial Biofilm Underneath the Wound Bed

Bacterial Biofilm in Wound Beds

The confocal laser scanning microscopy (CLSM) images depict the presence of Staphylococcus aureus (panels A and B) and Pseudomonas aeruginosa (panels C and D) within chronic wounds. The bacteria were identified using peptide nucleic acid fluorescence in situ hybridization (PNA-FISH) with species-specific probes: an FITC-labeled probe targeting S. aureus, which appears green, and a TxR-labeled probe targeting P. aeruginosa, which appears red. Arrows indicate the surfaces of the wounds, and the scale bar represents 30 micrometers.
